    Mechanism of Huiru decoction on hyperprolactinemia in rats

    Purpose: To investigate the anti-hyperPRL mechanism of traditional Chinese medicine Huiru Decoction (HD) in rats.Methods: The effect of HD on serum prolactin (PRL), estradiol (E2), progesterone (PGN), follicle stimulating hormone (FSH) and luteinizing hormone (LH) levels were investigated in hyperprolactinemia (hyperPRL) rats. Furthermore, the effect of HD on hypothalamus dopamine D2 receptor, protein kinase A (PKA) and cyclic adenosine monophosphate (cAMP) levels of hyperPRL rats were investigated respectively.Results: Compared with the model group, a high dose of HD (19.2 g/kg body weight) and medium dose of HD (9.6 g/kg body weight) reduced PRL level of hyperPRL rats effectively (p < 0.01). HD increased dopamine D2 receptor (p < 0.01), reduced cAMP (p < 0.01) and PKA (p < 0.01) levels of hypothalamus in hyperPRL rats significantly.Conclusions: HD showed anti-hyperPRL activity via dopamine D2 receptor.Keywords: hyperprolactinemia; Huiru Decoction; dopamine D2 recepto

    Population genetic structure and demographic history of small yellow croaker, Larimichthys polyactis (Bleeker, 1877), from coastal waters of China

    Small yellow croaker, Larimichthys polyactis (Bleeker, 1877), a commercially important benthopelagic fish, is widely distributed in the Bohai, Yellow and East China Seas. To evaluate the population genetic structure and demographic history of L. polyactis, we sequenced the complete mitochondrial deoxyribonucleic acid (mtDNA) control region (798 to 801 bp) in 127 individuals sampled from seven localities throughout its distribution region in China. A total of 136 polymorphic sites were detected, which defined 125 haplotypes. High haplotype diversity (1.000 ± 0.013 to 1.000 ± 0.034) and moderate nucleotide diversity (0.0112 ± 0.0061 to 0.0141 ± 0.0075) were detected in the species. The neighbor-joining tree of haplotypes was assigned into two closely related clades, but did not appear to have any geographic genealogic structure. Hierarchical molecular variance analysis (AMOVA), pair wise FST comparisons and the nearest-neighbor statistic (Snn) showed no significant genetic differences among populations in the Bohai, Yellow and East China Seas. The demographic history of L. polyactis was examined by using neutrality tests and mismatch distribution analysis, which revealed that the species had undergone a Pleistocene population expansion. The results based on the complete mtDNA control region sequences analysis indicate that within its distribution range, L. polyactis constituted a panmictic mtDNA gene pool. Factors such as dispersal capacity, ocean currents and insufficient evolution time could be responsible for the lack of population genetic differentiation in L. polyactis.Keywords: Larimichthys polyactis, mitochondrial control region, population genetic structure, demographi

    Effect of Eucommia ulmoides extract on osteoblast proliferation

    Purpose: To evaluate the effect of Eucommia ulmoides extract (EUE) on osteoblast proliferation as well as investigate its probable mechanisms of action.Methods: EUE was pharmacologically evaluated at three doses. Osteoblast cells were divided as follows: Group I: negative control; groups II–IV: received EUE (180, 360 and 540 μg/ml, respectively). We performed 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) assay to determine osteoblast viability following treatment. Alkaline phosphatase (ALP), osteocalcin, and collagen I levels in osteoblasts were quantified using commercially available kits. Thereafter, mRNA and protein expression of ALP, collagen I, osteocalcin, transforming growth factor-β1 (TGF-β1) were measured using real-time quantitative PCR (qPCR) and western blot, respectively.Results: EUE significantly (p < 0.01) promoted osteoblast proliferation at three treatment doses (180, 360, and 540 μg/mL). Furthermore, ALP, osteocalcin, collagen I and TGF-β1 expression at both mRNA and protein levels increased significantly (all p < 0.05) following EUE treatment.Conclusion: The results suggest that EUE may promote osteoblast cell proliferation and that ALP, osteocalcin, collagen I and TGF-β1 gene expression may be involved in the mechanism of action.Keywords: qPCR, collagen I, Bone, Liver, Eucommia ulmoides extrac

    Stock option, contract elements design and corporate innovation output – an analyse based on risk-taking and performance-based incentives

    Purpose: With the accelerated technological advancement, innovation has become a critical factor, which affects the core competitiveness of a company. However, studies about the relationship between internal stock option mechanisms and innovation productivity remain limited. Therefore, this paper aims to examine the impact of stock options and their elements design on innovation output from an internal mechanism perspective. Design/methodology/approach: Using a sample of 302 stock option incentive plans announced and implemented between 2006 and 2016, this study uses the propensity score matching and difference-in-difference model to find out whether the implementation of stock options improves the innovation outputs of enterprises. Findings: Based on the statistical analysis, it is concluded that: stock options can stimulate corporate innovation; a stock option may drive innovation outputs through two ways, performance-based incentives and risk-taking incentives, with the latter one playing a more dominant role and the risk-taking incentives of stock options, could be optimised when the non-executives granting proportion is larger, the granting range is limited, the incentive period is longer, the exercisable proportion is increasing, the price-to-strike ratio is lower and relatively loose performance assessment criteria are applied. Originality/value: The conclusion reached in the study may provide valuable information to listed firms in designing and implementing the stock option plans

    Total ginsenosides suppress monocrotaline-induced pulmonary hypertension in rats: involvement of nitric oxide and mitogen-activated protein kinase pathways

    AbstractBackgroundGinsenosides have been shown to exert beneficial pharmacological effects on the central nervous, cardiovascular, and endocrine systems. We sought to determine whether total ginsenosides (TG) inhibit monocrotaline (MCT)-induced pulmonary hypertension and to elucidate the underlying mechanism.MethodsMCT-intoxicated rats were treated with gradient doses of TG, with or without NG-nitro-l-arginine methyl ester. The levels of molecules involving the regulation of nitric oxide and mitogen-activated protein kinase pathways were determined.ResultsTG ameliorated MCT-induced pulmonary hypertension in a dose-dependent manner, as assessed by the right ventricular systolic pressure, the right ventricular hypertrophy index, and pulmonary arterial remodeling. Furthermore, TG increased the levels of pulmonary nitric oxide, endothelial nitric oxide synthase, and cyclic guanosine monophosphate. Lastly, TG increased mitogen-activated protein kinase phosphatase-1 expression and promoted the dephosphorylation of extracellular signal-regulated protein kinases 1/2, p38 mitogen-activated protein kinase, and c-Jun NH2-terminal kinase 1/2.ConclusionTG attenuates MCT-induced pulmonary hypertension, which may involve in part the regulation of nitric oxide and mitogen-activated protein kinase pathways
